Thanks I think that will work but before I try I'd like to ask one more question.

Suppose I have 2,500 directories on my NAS1 and each directory has 300 files totaling around 350MB.

On NAS2 I have an exact duplicate except I only have 2,499 directories. I want to copy just the one directory that is not on NAS2.

If I create a backup job as differential will it only copy the 1 directory and create 1 .bkf of just that directory? The directories on NAS2 are obviously not .bkf now. If it creates 1 massive .bkf the first time and then smaller ones after that, I don't have enough room or time to create the first massive backup.
